An precision pump moves the sample through the measuring pore in a measuring capillary. Here
is an electric field is applied. Each particle, passing the measuring pore, induces an
electric pulse, by displacing the buffer electrolytes. This individual pulse is measured as
“Count” .. Since each “Count” is actually created with a frequency of 1 million measurements per
second (1 MHz) ,, each particle is sized with a very accurate measurement of the
cell volume while for the whole sample a unique dynamic size range ranging from cellular
debris to large cellular aggregates is measured – in a single short measurement.
CASY takes to whole pulse area into account, precisely amplitude and pulse width. This enables a
high resolution measurement. Thus, even small changes in the membrane structure are visible: The
increased permeability of the membrane of dying or dead cells is detected reliably, until finally
only the remaining nucleus is detected.
The pulse area, namely, the combination of pulse amplitude and width is directly proportional to the volume of the
cell. This enables the determination of the individual cell volume and the
volume distribution of cells in the sample.
Yet another benefit to the user derives from the exact cell volume determination: The
CASY calculates the volumina of aggregates. Firstly, you get information about the
aggregation level of your sample and the exact biomass, and secondly, the volume can be
used to calculate the “true” cell number in the sample, by addition of the cells hidden in the aggregates,
thus taking all cells of your sample into account.
Zell debris can exert a disturbing influence on your cells and their behavior in scheduled
cell assays or influence the further use. The high dynamic measuring range of CASY
also captures cell debris and allows, within the usual 10 second measurement time, quick
assessment of the condition of your cell culture, and allows to optimize and standardize any costly cell assay
